Pentair Ultra Flow pump fails to prime. Hayward Vari Flo Valve 710XR50 ( unsure which handle location to dispell air? Thanks.

A pump should prime just fine on the filter setting but it will prime faster on recirculate. However, when a pump doesn't prime, it is usually because of either an impeller clog or a suction plumbing clog.
Did you recently vacuum a lot debris before this happened?
Remove the pump basket and check the impeller inlet for any debris. If that is clear, then try to back flush the suction line from the pump connection toward the skimmer using a hose wrapped in a wet rag.
